The HOPE-B trial evaluated health-related quality of life (HRQoL) in adults with severe or moderately severe hemophilia B two years after receiving etranacogene dezaparvovec gene therapy. Patient-reported outcomes indicated improvements in HRQoL domains such as treatment experience, emotional well-being, work/school responsibilities, and future outlook. However, physical functioning and pain showed limited improvement, reflecting irreversible joint damage accumulated before treatment.

These findings highlight the importance of early interventions, with emerging adolescent trials and gene-editing strategies like CRISPR-Cas9 offering potential for preventing joint disease in younger patients.
